CDK4 is a cyclin-dependent kinase regulator of cell cycle, found in all eumetazoans (animals other than sponges), and a regulator of G1. It is usually a single-copy gene, but has duplicated to give CDK4 and CDK6 in vertebrates. CDK4 kinases are regulated by Cyclin D, often in response to extracellular growth signals. CDK4 is also regulated by the chordate-specific INK4/CDKN genes. These are substantially expanded in vertebrates (p18/p19/p21/p27) as is cyclin D (cyclin D1/D2/D3). Retinoblastoma (Rb) is the best-known target of CDK4.

CDK4 is overexpressed or amplified in a number of human cancers, and is mutated in some rare familial cancers.
